Ginsenosides Re and Rg1 were transformed by recombinant β-glucosidase (Bgp1) to ginsenosides Rg2 and Rh1, respectively. The bgp1 gene consists of 2,496 bp encoding 831 amino acids which have homology to the glycosyl hydrolase families 3 protein domain. Using 0.1 mg enzyme ml−1 in 20 mM sodium phosphate buffer at 37°C and pH 7.0, the glucose moiety attached to the C-20 position of ginsenosides Re and Rg1, was removed: 1 mg ginsenoside Re ml−1 was transformed into 0.83 mg Rg2 ml−1 (100% molar conversion) after 2.5 h and 1 mg ginsenoside Rg1 ml−1 was transformed into 0.6 mg ginsenoside Rh1 ml−1 (78% molar conversion) in 15 min. Using Bgp1 enzyme, almost all initial ginsenosides Re and Rg1 were converted completely to ginsenosides Rg2 and RhThis is the first report of the conversion of ginsenoside Re to ginsenoside Rg2 and ginsenoside Rg1 to ginsenoside Rh1 using the recombinant β-glucosidase.

Ginsenosides are the principal components responsible for the pharmaceutical activities of ginseng. The minor ginsenosides, which are also pharmaceutically active, can be produced via the hydrolysis of the sugar moieties in the major ginsenosides using acid hydrolytic, heating, microbial, and enz...
Although the biotransformation of ginsenosides in the gastrointestinal tract has been extensively studied, much less is known about hepatic cytochrome P450 (P450)-catalyzed metabolism. The major aims of this study were to clarify the metabolic pathway and P450 isoforms involved and to explore the...
Rb1 and Rg1 are the major ginsenosides in protopanaxadiol and protopanaxatriol. Their content in ginsenosides was 23.8 and 17.6%, respectively. A total of 22 isolates of β-glucosidase producing microorganisms were isolated from the soil of a ginseng field using Esculin-R2A agar. Among these isol...

This study examined the transformation pathways of ginsenosides G-Rb1, G-Rb3, and G-Rc by the fungus Fusarium sacchari. Methods and Ginsenosides G-Rb1, G-Rb3 and G-Rc were isolated from leaves of Radix notoginseng, and their structural identification was confirmed using NMR. Transformation of G-Rb...
Ginsenosides such as Rb1, Rg3 and Rh2 are major bioactive components of Panax ginseng. This in vivo study investigates the metabolic pathways of ginsenosides Rb1, Rg3 and Rh2 orally administered to rats. High performance liquid chromatography-mass spectrometry (LC-MS) and tandem mass spectrometry...
20(S)-Protopanaxadiol (PPD, 1) is one of the aglycones of the ginsenosides and has a wide range of pharmacological activities. At present, PPD has progressed to early clinical trials as an antidepressant. In this study, its fate in mixed human liver microsomes (HLMs) and human hepatocytes was exa...
Baker's yeast (Saccharomyces cerevisiae) whole-cell bioconversions of naringenin 7-O-β-glucoside revealed considerable β-glucosidase activity, which impairs any strategy to generate or modify flavonoid glucosides in yeast transformants. Up to 10 putative glycoside hydrolases annotated i...
Fungi are used in many industrial processes, such as the production of enzymes, vitamins, polysaccharides, polyhydric alcohols, pigments, lipids, and glycolipids. Some of these products are produced commercially while others are potentially valuable in biotechnology. Fungal secondary metabolites ...
Hydroxycinnamic acids are a class of phenolic antioxidants found widely in dietary plants. Their biotransformation in the human organism primarily involves Phase II conjugation reactions. In this study, activities of UDP-glucuronosyltransferases (UGTs) and sulfotransferases (SULTs) towards major ...
The product spectrum of a soil bacterium Corynebacterium glutamicum was extended to include a functional sugar xylitol. The recombinant C. glutamicum, engineered to express the xylose reductase gene XYL1of Pichia stipitis, produced xylose reductase with a specific activity of ca. 0.6 U/mg protein...
